No prob - thanks for confirming, and sorry I couldn't provide anything more concrete. I've found very rarely, like this, something will glitch out with an instance which isn't easily explained by anything we can see on the development side. But luckily I've found this is pretty rare.

Great! I started a new instance and the correct user is now set as the assignee.

Everything looks ok so far - other than the stuff I mentioned I'm out of ideas for the moment. Perhaps launch a new instance of the task and see if you can assign that? Otherwise I'd suggest checking the tomcat STDOUT logs to see whether it provides any more specific error messages, etc.

Resume Process is greyed out, but everything else is available. I should add that this process has run successfully hundreds of times before this problem

How about if you click the "modify" dropdown in the Process Instance window frame? Are "Pause" and "Cancel" un-greyed-out?

It sounds like there might be some deeper issue with your process instance - like an actual underlying error either with the task or with the process instance. Could you post a screenshot of what you see when you right-click the task and click "instances"?
