After importing all the certificates (including intermediate ones) in the keystore the email Integration has started working. Importing the certificate in the appian admin site was not enough.

Does the certificate still currently exist in your java keystore (I'm assuming cacerts file in JAVA_HOME)?
On a previous upgrade from 17.4 to 18.4 we were able to add locally signed cert in the admin console but had to remove them from the java keystore to get it to work for calling webservices.
