You'd probably be interested in the function isUsernameAvailableForNewAccount() found in the Personalization Utilities plug-in. Beyond that, if some usernames in your system will be hand-entered by other users, I strongly suggest forcing them to either all lowercase or all uppercase, to remove some of the guesswork.

How do you plan to create users? In admin console you can not check that. Using a process it should be simple to just validate against existing users and might need a combination of OOTB functions and plugins. If you do that through LDAP or SSO this is managed elsewhere.
