Regardless of any layout component. A file upload will only work in processes. Mike already mentioned this.

If you want to stay with an interface in the site, then you can use a startProcessLink to upload files using a process.

I'm not clear what this interface is attempting to do, but if it's not in a user input task or start form of a process, the File Upload Field will not be functional.

Actually, I have a side navigation view on my interface where in when I select an option from left side menu, I get to see related content (interface) on the right side, Now the problem I am facing here is one of my content (interface) has fileupload component in it, if I call that interface without form the fileupload component turns disabled and if I use form layout in that interface I can not call the interface in content area (which is column layout). Please help me with it.

Why would you want to do this?

Either way, the answer is "no" - a!formLayout() must be the top-level interface object (when used at all) in the interface for a user input task / start form. It's also not required to be used in either of these, it just makes certain things easier. And it makes no difference to whether the File Upload Field will work.
