How can we set a link which is valid for 2 hours only? And I want to send it by email object to someone. If someone tries to open this link after 2 hours it will display a message only e.g. Already submitted!!

Why not assign a task to this person and add an exception for 2 hours.
Who is "someone"? Is this an Appian user?

If the user is not an Appian user, then I assume that the link also does not belong to Appian. In that case, you will have to add the security on your destination (i.e. where the link is taking you)
I am not sure I understand your question.
A non Appian user cannot access the system. This would be a use case for Appian Portals. And your use case is totally supported using portals.
