We would like to trigger a Process by an email. For this we are planning to add Alias - abc@domain.com with UUID in custom.properties but we don't want to add any KEY (DestinationPMUUID) anywhere in Email. Is Alias and UUID at custom.properties only need to trigger process or do we need to add KEY (DestinationPMUUID) also in EMAIL ( Subject, Message Body etc..)

As long as the alias is defined and points to a valid process model, process or event, it is not required to include the DestinationPMUUID key-value pair in the subject line in the email. Refer to the documentation for setting up routing aliases -
forum.appian.com/.../Mail_Server_Setup.html
under sub-section titled "Adding a Routing Alias to custom.properties"
