We are using the Hemberger icon with large menu. We should be able view menu on-mouse over, How we can achieve this in appian ?

Appian does not support dynamic UI on mouse-over.

Not recommended, but if this is a must have feature, then, yes you can develop custom component plugins.

Appian is about low-code. Not an application server to run your custom JS stuff. Component plugins are a way to add functionality which would be a deal-breaker for your project.

You can not extend an existing component if this is what you mean.
Your component needs to implement the whole visual display and behaviour.
But, in the first place, try to use OOTB components.
Agreed with Stefan - can you provide more context on what the purpose of this user interface is? There's a ton of ways that can likely meet your requirements even if it doesn't display exactly using a hover menu.
